Abstract

Source/description: The chicken luteinizing hormone/choriogonadotropin receptor gene has been cloned and sequenced. To produce markers for linkage mapping genomic DNA sequence from exon 7 to exon 9 including the intervening introns and exon was cloned and sequenced (EMBL accession number AJ289775). The intron-exon structure of this region, including exon 8 was the same as in mammalian LHCGR genes. A pair of primers for PCR which gave a single amplification product of 287 bp were then designed for the purposes of mapping. The sequence of the amplified product was confirmed by sequencing (97.6% sequence identity with AJ289775). Polymorphism was revealed by single strand conformation polymorphism (SSCP) analysis.

Chromosomal location: Segregation analysis indicated that the chicken luteinizing hormone/choriogonadotropin receptor gene was located on chromosome 3, placing it at position 45.35. The marker for the luteinizing hormone receptor gene (ROS0250) is tightly linked (phi = 0.0) to MCW0169 and ADL0177 with lod scores of 14.4 and 15.7, respectively. There was no evidence for conservation of gene order in this region of chicken chromosome 3 with human or mouse chromosomes.
