Abstract

Source/description: Identification and analysis of chromosomal regions which are orthologous in different species of warm-blooded animals is the basis of comparative genome mapping. In addition, knowledge of orthology between functionally specialized chromosomal regions in diverse mammalian and avian genomes could aid in clarifying common features of genome organization in these species. Because of a high degree of evolutionary conservation, the coding sequences are preferable for comparative mapping. Functional specialization of chromosomal regions is reflected by their compositional heterogeneity, presence of CpG islands, and NotI restriction sites. The NotI linking clones contain sequences that flank a single NotI recognition site, while NotI jumping clones contain DNA sequences that lie between neighbouring NotI restriction sites. Such clones have been shown to be associated with CpG islands and genes. Three genomic NotI linking libraries with high gene density have been established for human chromosome 3. Contigs of NotI clones that span HSA 3p14–21 and HSA 3q13–23 have been established by fluorescence in situ hybridization (FISH)- and radiation hybrid (RH)-mapping.
